K100D กล้องดิจิตอล SLR รุ่นใหม่จาก PENTAX มีคุณสมบัติที่โดดเด่นคือระบบป้องกันภาพสั่นไหว Shake Reduction System โครงสร้างภายในเป็นโลหะสแตนเลส ชุดวงจรเซอร์กิตเป็นแบบมัลติ-เลเยอร์ ตัวกล้องขนาดเล็ก น้ำหนักเบา ระบบบันทึกภาพออกแบบให้ใช้งานง่ายเหมาะสําหรับผู้เริ่มต้นถึงระดับมืออาชีพ
K100D ยังคงใช้เซ็นเซอร์ภาพ CCD ขนาด 23.5 x 15.7 มม.ความละเอียด 6.1 ล้านพิกเซล เลือกปรับระดับการบีบอัดข้อมูลได้ 3 ระดับคือ Best, Better และ Good ไฟล์ฟอร์แมทในตัวกล้องมีทั้งแบบ RAW และ JPEG ให้เลือกใช้ ระบบการประมวลผลภาพแบบใหม่ ให้สีสันภาพสมจริง มีการไล่เฉดสีที่ถูกต้องแม่นยํา ช่องมองภาพเป็นแบบมองผ่านเลนส์ TTL ใช้กระจกสะท้อนภาพแบบแพนทาไมเรอร์ ให้ความสว่างสูง มองเห็นได้เหมือนจริง 96% และมีระบบกันภาพสั่นไหวแบบใหม่ในตัวกล้อง Shake Reduction (SR) ทำให้ถ่ายภาพในสภาพแสงน้อยได้โดยไม่ต้องกังวลเรื่องภาพเบลอ รวมไปถึงการใช้เลนส์เทเล หรือเลนส์มาโคร และการใช้แฟลชด้วยความ เร็วชัตเตอร์ต่ำ สามารถบันทึกภาพได้ดีกว่ากล้องทั่วๆ ไปที่ไม่มีระบบป้องกันภาพสั่นไหว สามารถใช้ได้กับเลนส์ทุกตัว

เม้าท์เลนส์เป็นแบบ K-, KA-, KAF 2 สามารถใช้ร่วมกับเม้าท์เลนส์กล้องขนาดกลาง 645 และ 67 โดยใช้งานร่วมกับอะแดปเตอร์ (ใช้ได้ในบางฟังก์ชั่น) สำหรับเลนส์กล้อง 35 มม. ต้องคูณทางยาวโฟกัส 1.5 เท่า
ระบบออโต้โฟกัสใหม่ ทำงานด้วยความเร็วสูง วางตําแหน่ง 11 จุดกระจายทั่วทั้งจอภาพ ทำให้ปรับโฟกัสได้อย่างถูกต้องไม่ว่าวัตถุจะอยู่ในส่วนใดของภาพ โดยใช้เซ็นเซอร์แบบ SAFOX VIII AF เลือกจุดโฟกัสหรือเลือกปรับเองได้ มีระบบโฟกัสแบบทีละภาพและแบบต่อเนื่องตามการเคลื่อนไหว
ระบบวัดแสงเป็นแบบแบ่งพื้นที่ 16 ส่วน หรือเลือกแบบมัลติแพทเทิร์น เฉลี่ยหนัก กลาง และแบบเฉพาะจุด เลือกปรับชดเชยแสงได้ +/-2 สตอป ปรับขั้นละ 1/2, 1/3 สตอป ล็อคความจําแสงได้ ช่วงความเร็ว ชัตเตอร์ที่ 30-1/4000 วินาที และมีชัตเตอร์ B สําหรับการถ่ายภาพที่ต้องใช้ระยะเวลานานกว่า 30 วินาที และมีระบบหน่วงเวลา 2 หรือ 12 วินาที มีฟังก์ชั่นถ่ายภาพต่อเนื่องได้ที่ความเร็ว 2.8 เฟรม/วินาที ต่อเนื่องกัน 5 เฟรม/วินาที (ไฟล์ JPEG) และ 3 เฟรม/วินาที (ไฟล์ RAW) ระบบไวท์บาลานซ์เป็นแบบออโต้ เลือกให้เหมาะสมตามสภาพแสงได้ เช่น แสงกลางวัน, แสงในที่ร่ม, มีเมฆมาก, ไฟทังสเตน, ไฟฟลูออเรสเซ้นท์และไฟแฟลช สามารถเลือกปรับตั้งเองแบบแมนนวลได้ ความไวแสงเป็นแบบออโต้ ISO 200-1600 ปรับเลือกได้สูงสุดถึง ISO 3200 มีฟังก์ชั่นลด Noise เมื่อถ่ายภาพเป็นเวลานาน หรือบันทึกภาพด้วยความไวแสงสูง และมีระบบถ่ายภาพคร่อมอัตโนมัติ

K100D มีระบบบันทึกภาพเพื่อการใช้งานง่ายๆ แบบ Auto Picture กล้องจะเลือกระบบบันทึกภาพสําเร็จรูปให้อัตโนมัติ ซึ่งกล้องจะคํานวณการเปิดปิดรูรับแสงให้พอดีขึ้นอยู่กับสภาพแสงและสิ่งที่ต้องการจะถ่าย หากแสงน้อยเกินไปแฟลชในตัวแบบป๊อปอัพก็จะทํางานโดยอัตโนมัติ และมีโปรแกรมมาตรฐาน ให้เลือกใช้อย่างครบครันไม่ว่าจะเป็น P, S, A, M ซึ่งเหมาะสำหรับนักถ่ายภาพที่ต้องการปรับตั้งการทำงานด้วย ตนเอง รวมทั้งโปรแกรมถ่ายภาพสําเร็จรูปมีให้เลือกถึง 7 แบบ ได้แก่ ถ่ายภาพทั่วไป, บุคคล, ทิวทัศน์, มาโคร, บุคคลกลางคืน, กีฬาและภาพไม่ใช้แฟลช

ทางด้านหลังมีจอมอนิเตอร์ LCD ขนาดใหญ่ 2.5 นิ้ว แบบ TFT ความละเอียด 210,000 พิกเซล มองเห็นได้ชัดเจนเต็มเฟรม เลือกปรับซูมดูภาพได้มากถึง 12 เท่า ด้านบนเป็นแฟลชแบบป๊อปอัพขนาดเล็ก ทํางานในระบบ P-TTL มีค่าไกด์นัมเบอร์ 15.6 รองรับการใช้เลนส์มุมกว้างถึง 20 มม. ความเร็วชัตเตอร์สัมพันธ์แฟลชที่ 1/180 วินาที หรือต่ำกว่า พร้อมกับฮอทชูสําหรับใช้งานร่วมกับแฟลชภายนอก

คุณสมบัติอื่นที่น่าสนใจเช่น มีคัสตอมฟังก์ชั่นให้เลือกถึง 19 แบบ สําหรับปรับตั้งการทํางานแบบต่างๆ เลือกปรับแต่งภาพให้สวยงามได้ 5 ระดับ ใน 3 รูปแบบ คือ ความอิ่มตัวของสี คอนทราสท์ และความคมชัด และยังมีฟังก์ชั่นฟิลเตอร์ดิจิตอลในตัว อาทิ ฟิลเตอร์ ซอฟท์ช่วยให้ภาพดูนุ่มนวล, ฟิลเตอร์แบบขาวดํา-ซีเปีย และฟิลเตอร์สลิม ช่วยให้ภาพดูแคบลงกว่าปกติ ทางด้านการพิมพ์ภาพโดยตรงรองรับการพิมพ์ตามมาตรฐาน PictBridge การเชื่อมต่อภายนอกมีอินเตอร์เฟส USB 2.0 แบบ Hi-Speed ทำให้โหลดภาพได้อย่างรวดเร็ว การจัดเก็บภาพใช้เมมโมรี่แบบ SD Card เลือกใช้แบตเตอรี่ได้ 2 ชนิดคือ ลิเธี่ยม CR-V3 สองก้อนหรือแบตเตอรี่ AA 4 ก้อน จะใช้แบบอัลคาไลน์หรือแบตเตอรี่ชาร์จ NiMH ก็ได้
